The Repair Champion in High-Temperature Environments! T&L Biotechnology Officially Launches New-Generation Thermostable bFGF Protein
Basic Fibroblast Growth Factor (bFGF), a member of the fibroblast growth factor family, triggers signaling transduction pathways by binding to and activating receptors on targeT Cell surfaces. bFGF regulates the proliferation of diverse cell types, including endothelial cells, neurons, chondrocytes, smooth muscle cells, melanocytes, and adipocytes. Additionally, bFGF is involved in biological processes such as cell self-renewal, differentiation, development, tissue repair, and regeneration. As a critical factor for cell growth, bFGF is widely used in the in vitro culture of various cell types.
Studies show that bFGF is unstable under physiological conditions, with a half-life of approximately 8 hours in mammalian cell cultures. The low thermal stability and short half-life of bFGF lead to challenges in cell culture, requiring frequent growth factor supplementation or daily medium replacement. The thermostable bFGF protein, engineered through modification of natural bFGF, addresses this challenge. It retains the potent biological activity of natural bFGF while achieving enhanced thermal stability, maintaining structural integrity and protein activity across a broad temperature range.
Human embryonic Stem Cells (hESCs) and induced pluripotent stem cells (iPSCs) possess the potential to differentiate into all somatic cell types. During in vitro culture of hESCs and iPSCs, bFGF is essential for maintaining stem cell pluripotency. Research demonstrates that hESCs cultured with thermostable bFGF form undifferentiated colonies faster (Fig. 1A) and exhibit enhanced proliferation rates during long-term culture (Fig. 1B). Further studies reveal that thermostable bFGF promotes hESC proliferation while sustaining the expression of pluripotency markers (e.g., OCT4, SOX2, KLF4), validating its ability to preserve hESC pluripotency—a critical feature for cell culture applications.

When added to iPSC culture medium, medium replacement can be performed every 2 days.
As shown in Fig. 5, T&L’s Recombinant Human bFGF Protein (GMP-TL901) and Thermostable bFGF were applied to AMMS® Pluripotent Stem Cell Medium (AS-35) for iPSC culture. In medium supplemented with thermostable bFGF, daily or every-other-day medium changes significantly improved colony numbers and sizes compared to regular bFGF. Notably, cell viability showed no significant difference between daily and every-other-day protocols with thermostable bFGF, validating its suitability for cost-efficient iPSC culture.
