T cells are an elite force within our immune system, and T cell therapy is demonstrating significant potential in tackling challenging diseases like cancer and autoimmune disorders, offering new treatment options for more patients. If the activation intensity is too low, T cell transduction efficiency may decrease, impairing its antitumor effect. Typically, the CAR-T process includes steps such as patient blood collection, T cell isolation, T cell activation, transduction, expansion, and reinfusion. Among these, T cell activation is a critical step that not only influences subsequent cell proliferation but also directly affects the efficacy and persistence of CAR-T cell therapy. One of the most widely used strategies for T cell activation is to use CD3/CD28 antibody-coupled microbeads or nanoparticles, providing the necessary first and second signals required for T cell activation and proliferation. Nanoparticle-based reagents, with their ultra-fine particle size, remain evenly dispersed in the culture medium without sedimentation, offering uniform contact and activation of T cells, and require no pre-washing, making them easy to use. These advantages make nano-based T cell activation reagents an attractive option for CAR-T therapy.
